چکیده

supplemented with 10% horse serum. The cells grew ex ponentially with a doubling time of 16 to 17 hours, had a cell volume (mean ±S.E.) of 2313 ±89 cu /tm, and were cloned in soft agar with a cloning efficiency of 68.7 ±6.3%. Sensitivity to the alkylating agent nitrogen mustard (HN2) was comparable to that observed previously with LSI78Y murine lymphoblasts; the D0 (the dose of drug reducing survival to 37% of the initial cell population) was 5.35 ng/ml, and the extrapolation number, n, was 1.27. An investigation of the interaction of hydrolyzed nitrogen mustard and choline transport revealed that uptake of choline-14C obeyed simple Michaelis-Menten kinetics, proceeded "uphill" against a concentration gradient of over 30-fold, and showed a decrease in distribution ratio as choline concentration increased and choline transport was competitively inhibited by hydrolyzed HN2. The Michaelis constant, Km, for choline transport was 2.90 x 10~5M,the transport capacity, Vmttx, was 4.83 x 10~17 moles/min/

Nucleoside transport in Walker 256 rat carcinosarcoma and S49 mouse lymphoma cells. Differences in sensitivity to nitrobenzylthioinosine and thiol reagents.

The characteristics of nucleoside transport were examined in Walker 256 rat carcinosarcoma and S49 mouse lymphoma cells. In Walker 256 cells the initial rates of uridine, thymidine and adenosine uptake were insensitive to the nucleoside transport inhibitor nitrobenzylthioinosine (NBMPR) (1 microM), but were partially inhibited by dipyridamole (10 microM), another inhibitor of nucleoside transpo...

Drug-induced Stimulation of Nitrogen Mustard and Choline Transport and Other Systems in L5178Y Lymphoblasts in Vitro1

The effect of 5 drugs on the transport of several substrates, including the chemotherapeutic agent nitrogen mustard; choline; two nonmetabolizable amino acids, a-aminoisobutyric acid and cycloleucine; the glucose analog, 3-0-methyl-Dglucose; and the heme precursor, 6-aminolevulinic acid; was undertaken in L5178Y murine lymphoblasts in vitro. Trans port of both hydrolyzed nitrogen mustard and ch...

Sensitivity to nitrogen mustard as a function of transport activity and proliferative rate in L5178Y lymphoblasts.

natural substrate choline was also a function of the proliferative state of the cells. Transport was more efficient in log-phase cells as manifested by a higher binding affinity between carrier and each of the three substrates; in addition transport capacity for hydrolyzed HN2 and choline was significantly greater in log-phase cells than resting cells. The greater sensitivity of log-phase cells...

Protection of Ehrlich ascites tumor cells against the antiproliferative effect of mechlorethamine (nitrogen mustard) by 5-N,N-dimethylamiloride.

5-N,N-Dimethylamiloride protects Ehrlich ascites tumor cells against the antiproliferative effect of nitrogen mustard. The drug reduces the frequency of DNA interstrand cross-links introduced by nitrogen mustard. Cells with a defective choline carrier are not protected against nitrogen mustard by dimethylamiloride. As nitrogen mustard is taken up by the choline carrier, it is concluded that the...
